ANTIBACTERIAL ACTIVITY OF LENTINULA EDODES GROWN IN LIQUID MEDIUM BJM

The antibacterial activity of 35 isolates of Lentinula edodes, a shiitake mushroom, against Bacillus subtilis was evaluated by diffusion technique in agar with a semi-solid overlay. All isolates inhibited B. subtilis and the isolate Le1 promoted the formation of the largest inhibition zone. L. edodes Le1 also presented antibacterial activity against foodborne pathogens and food contaminant bacteria, particularly Gram-positive species. The antibacterial activity of the culture filtrate after 18-25 days of cultivation of L. edodes in broth at 25ºC was high. The inhibitory activity was observed only in the organic layer when the culture filtrate was partitioned between ethyl acetate and water, suggesting that the inhibitory substances have low polarity. The antimutagenic effect of the mushrooms Lentinula edodes and Agaricus blazei was studied on conidia of Aspergillus nidulans when exposed to short wave ultraviolet light. Two strains of A. nidulans were used. For the preparation of the extracts, the fresh mushrooms were left in aqueous infusion for 12 hours and heated in a water bath for 15 min at 100ºC, and then the material was filtered. The dehydrated mushrooms were left in aqueous infusion for 12 hours and to filtrated. Both filtrates were used as extracts. Avaliou-se o crescimento micelial das linhagens de shiitake LE-95/01, LE- 96/18, LE-96/13, LE-98/55, LE-98/47, LE-95/02, LE-95/06 e LE-95/07, a sua produção em toras de eucalipto e as conseqüentes alterações em características físicas da madeira em decorrência da inoculação com a linhagem LE-96/18. O crescimento micelial foi avaliado em meio de cultura SA (serragem enriquecida com farelo-ágar) a 25ºC. No cultivo em toras, cada linhagem foi inoculada em 40 toras de Eucalyptus grandis com 1 m de comprimento e 9 a 14 cm de diâmetro. The mycelium growth kinetics and vigor of shiitake (Lentinula edodes (Berk.) Pegler) strains LE 96/17, LE 98/51, LE 98/53, and LE 98/56 were studied under different agar medium compositions. The strains were from the mycological collection of the Módulo de Cogumelos, Faculdade de Ciências Agronômicas, Unesp-Botucatu, Brazil. Effect of Agaricus brasiliensis and Lentinula edodes mushrooms on the infection of passionflower with Cowpea aphid-borne mosaic virus BABT

The objective of the present study was to evaluate the protection of passion fruit plants against CABMV by using preparations from Agaricus brasiliensis and Lentinula edodes mushrooms. In experiments carried out in the greenhouse, the fruiting body extracts from some of the isolates of both mushrooms significantly reduced CABMV incidence in passion fruit plants. This protective effect occurred when the plant leaves, pre-treated with extracts, were later inoculated mechanically with the virus. However, the extracts did not protect the plants in experiments involving CABMV transmission by aphid vectors. Growth and antibacterial activity of Lentinula edodes in liquid media supplemented with agricultural wastes Electron. J. Biotechnol.

Antibacterial activity of Lentinula edodes against Bacillus subtilis was evaluated in cell-free filtrates obtained after growth in 14 different culture media. The highest B. subtilis growth inhibition was promoted by filtrates of growth media supplemented with rice bran, vermiculite or molasses. L. edodes dry mycelial biomass in liquid culture with 0.5% added rice bran was 3.2 mg/ml, after growth for 30 days at 25ºC without shaking, and 4.3 mg/ml under orbital shaking (150 rpm). However, antibacterial activity, detected between 20 and 24 days of incubation of stationary cultures, was absent in filtrates of aerated cultures. Influence of stationary and bioreactor cultivation on Lentinula edodes (berk) pegler lignocellulolitic activity BABT

This work aimed to study the stationary and periodically mixed culture of L. edodes to the production of lignocellulolitic enzymes activity. LE 95/17, LE 96/22 and Leax strains were incubated in 25 g of eucalyptus sawdust substrate in Erlenmeyer flasks in stationary culture at 25º C and in a bioreactor with four complete rotations daily at 25º C and 3% CO2. The samples were collected at 8, 11, 14, 17 and 20 days after the incubation. Oxidative and hydrolytic enzymes analyses were performed. Obtenção de linhagens de Lentinula edodes resistentes a temperaturas elevadas e seleção de linhagens resistentes ao Trichoderma sp Ciência e Agrotecnologia

Lentinula edodes (Berk.) Pegler (shiitake) é o segundo cogumelo mais consumido no mundo e sua produção pode ser feita em toras de madeira ou em cultivo axênico utilizando resíduos agroindustriais como substrato. O cultivo do shiitake no Brasil tem sido gravemente prejudicado pela alta incidência de fungos contaminantes, como Trichoderma sp., e pela ausência de linhagens mais adaptadas às temperaturas tropicais. Duas linhagens resistentes ao Trichoderma sp. foram obtidas por meio de seleção quando crescidas em contato com o fungo contaminante e, por meio de fragmentação micelial, foram obtidas linhagens resistentes a temperaturas elevadas, capazes de crescer a 30º C em meio de cultura.

Partial purification of elicitors from Lentinula edodes basidiocarps protecting cucumber seedlings against Colletotrichum lagenarium BJM

The shiitake mushroom (Lentinula edodes) has been used in research involving the prevention and the control of human and plant diseases. In cucumber plants, treated with aqueous extracts from mushroom basidiocarps, there was a reduction in anthracnose severity caused by Colletotrichum lagenarium, and an increase in peroxidase activity in the leaves. With the aim of obtaining molecules of agronomic interest, the crude aqueous extract from L. edodes basidiocarp was fractioned with ammonium sulfate. Laccases are enzymes involved in lignin degradation and are produced by various organisms. Due to their low substrate specificity their potential to be used in biotechnological applications has received attention. The addition of laccase inducers to the culture medium of microorganisms can enhance laccase production and facilitate its purification and utilization. This investigation was performed to evaluate the biological efficiency (BE), mean mushroom weight (MMW), mean number of mushroom (MNM) and mushroom quality of Shiitake [ Lentinula edodes (Berk.) Pegler] when grown on a sterilized substrate composed by sugarcane bagasse enriched with rice bran and sugarcane molasses. Strains of Lentinula edodes suppress growth of phytopathogenic fungi and inhibit Alagoas serotype of vesicular stomatitis virus BJM

Four Lentinula edodes strains (Le10, 46, K2, Assai) were assessed for their antagonistic effect on four filamentous fungus species of agricultural importance (Helminthosporium euphorbiae, Helminthosporium sp, Fusarium solani and Phomopsis sojae) and on Alagoas serotype of Vesicular Stomatitis Virus (VSA). The L. edodes strains studied had variable effects on the filamentous fungi and on VSA. The K2 and Le10 strains were antagonistic on the fungi assessed and the 46 and K2 strains were efficient on the Vesicular Stomatitis Virus. The results widened the list of beneficial effects of L. edodes on the control and prevention of animal pathogenic virus and filamentous fungi.

Supplementation of sugarcane bagasse with rice bran and sugarcane molasses for shiitake (Lentinula edodes) spawn production BJM

The purpose of this study was to assess the myceliation rate, mycelial vigor and "estimated biomass" of Lentinula edodes (Berk.) Pegler, grown on a sugarcane bagasse substrate enriched with rice bran and sugarcane molasses for spawn production. The proportions of rice bran used were 0, 10, 15, 20, 25, 30 and 40% (dry weight/dry weight of bagasse) and the sugarcane molasses concentrations tested were 0, 10, 20, 30, 40, 50 and 60 g/kg (dry weight/dry weight of bagasse plus rice bran). Thermal and mechanical shocks affecting the first flush of production of Lentinula edodes on Eucalyptus saligna logs BJM

The aim of this work was to evaluate the effect of thermal and mechanical shocks on the productivity of Lentinula edodes colonized on 140 Eucalyptus saligna logs at different immersion times in water and in the first flush of production. The logs were immersed in water at low temperature (16ºC) or environmental temperature (22ºC), for 6, 10, 14, 18, 22, 26 and 38 hours. The mechanical shock treatment was accomplished by dropping the logs onto the floor three consecutive times, in a vertical position. Water temperature and immersion time affected L. edodes yield, which increased two to four times when the logs were immersed in cool water for the shortest times (6 and 10 hours). The mechanical shock treatment did not increase the sporophore yield.

Viabilidade econômica do cultivo de shiitake em diferentes escalas de produção Scientia Agricola

O interesse nacional pelo cultivo comercial de shiitake é crescente, devido às suas propriedades nutricionais e medicinais, e ao seu apreciável sabor. O cultivo deste cogumelo constitui uma alternativa rentável de diversificação das atividades de uma propriedade agrícola. Este trabalho teve como objetivo determinar, no Estado de São Paulo, a viabilidade econômica do cultivo de shiitake considerando-se um ciclo de 14 meses, em 1000, 2000, 3000 e 4000 toros de eucalipto.
